HEPES Buffer in Detail

HEPES is a Good's buffer reagent, the full name is N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid. Its pH buffer range is in the range of 6.8 to 8.2. It is easily soluble in water, and there is no precipitation reaction with multivalent metal ions. In addition, most of the natural factors will not affect the dissociation of HEPES, such as temperature, concentration, medium salt and ion composition, etc. It can be seen that HEPES buffer is very stable.

Applications of HEPES

How to use HEPES Buffer:

The first method of use is that HEPES powder is directly added to the prepared culture solution according to a certain concentration required, and then filtered and sterilized. Add 2.38g of HEPES buffer reagent to each thousand milliliters of culture solution, and adjust the pH to 7.2 with 1N sodium hydroxide after dissolving, and then use it normally after filtration and sterilization. At this time, the concentration of HEPES biological buffer is 10 mmol/L.

The second method of hepes buffer use usually takes 99 milliliters of culture solution before use, and then adds 1 milliliter of stock solution. At this time, the application concentration of this solution is still 10 mmol/L. Dissolve 23.8g of HEPES in 90ml of double-distilled water, adjust the pH to 7.5 to 8.0 with 1N sodium hydroxide, start to dilute to 100ml with water, filter and sterilize, divide into 2ml vials, and store at 4°C or -20 °C. This is how a 1 mol/L (100 x) HEPES stock solution is formulated.
