Characteristics and Use of Good's Buffer PIPES

PIPES is a member of the ethanesulfonic acid buffer series, known as 1-4-piperazine diethyl sulfonic acid in Chinese. It is one of the ethanesulfonic acid buffer reagents developed by Good et al. in 1960, and has some important characteristics common to Good's buffer solutions, such as medium pKa, maximum water solubility, and minimum solubility in all other solvents, minimum salt effect, minimum pKa with temperature variability, chemical and enzymatic stability (PIPES is insoluble in water) Minimal absorption of visible or ultraviolet spectral range, easy to synthesize, and other characteristics.

The effective buffering range of PIPES is pH 6.1-7.5, with pKa=6.8 (25 ℃). Its pKa is close to physiological pH and suitable for cell culture. In addition, PIPES can also serve as a buffer reagent in animal and plant chemistry to minimize lipid damage and experimental artifacts caused by glutaraldehyde fixation. It is worth noting that PIPES is not suitable for use as an oxidizing reducing agent due to its ability to form free radicals.

PIPES can be used for in vitro cell culture medium. In the study of measuring lactate in plasma using lactate oxidase method, it was found that PIPES can stabilize for 172 hours at 37 ℃. 4% paraformaldehyde prepared with 50mmol/L PIPES buffer can be used for fixation during immunofluorescence labeling. PIPES can meet the requirement of extremely low complexation of divalent cations when adenosine diphosphate (ADP) binds to relaxed fan-shaped myofibroblasts.
